International Residential Code requires at least 21 inches of clear floor space in front of the sink and vanity, even in a small bathroom. Start by measuring the front edge of the wall or fixture opposite your existing sink or vanity (or the spot where you want the new one if you're doing a full remodel). You'll need to make sure you leave at least 15 inches of clear floor space when you measure from the center of the vanity to any side wall. And if you have family members with mobility issues, you'll want to increase those measurements. For example, in public spaces, the Americans with Disabilities Act requires 30 inches in front of bathroom vanities and 48 inches of side clearance.
